Coliform and E. coli

Coliforms in general indicates the presence of faecal contamination. Coliforms do not always cause illness in humans but they indicate the presence of other disease-causing organisms like typhoid, dysentery. Both these types of bacteria live in water but disease-causing bacteria often do not live long and this makes detection difficult. Coliforms on the other hand last much longer. However, we must know that there are two real sources coliforms; one from animal intestinal tract and the other from soil vegetation and insects. Non faecal coliforms are Enterobacter, Klebsiella and Citrobacter. As such faecal coliform which is usually found in the guts of warm-blooded animals have been the choice indicator organism for faecal contamination. However, in 1986 the USEPA recommended that E. coli be used as an indicator of faecal contamination instead of E. coli based on an epidemiological survey where E. coli proved to have a better correlation to swimming illness than faecal coliform. E. coli is rod shaped and is different from the coliforms by its ability to ferment lactose at 44°C. When cultured on eosin methylene blue green metallic colonies on a dark purple media indicates the presence of E. coli. Incubation is however up to 48-72 hrs under the optimal growth temperature of 37°C. Mos strains of E. coli are harmless but some cause serious symptoms like bloody diarrhoea, stomach cramps and vomiting.

Film Method - Compact Dry EC

R-Biopharm Indonesia supplies the Compact Dry EC. The ready-to-use plates consist of a special 50 mm diameter petri dish containing a detection specific nutrient pad. The nutrient medium of Compact Dry EC contains two chromogenic enzyme substrates: Magenta-Gal and X-Gluc. Thus, coliforms show a red or red violet coloration of their colonies while colonies of E. coli will appear in blue. By addition of all red, red violet and blue colony counts the total number of coliform bacteria grown on the nutrient can be obtained.

Merck Biocontrol Indonesia has two platform for the detection of E. coli and coliforms. One is coli complete which uses a supporting substrate disc method to detect the presence of beta galactosidase produced by coliforms. E. coli fluoresce under UV light by the reaction of their enzyme beta glucuronidase. Another platform is the Simplate. This technology uses a proprietary Binary Detection technology to provide quantitative results for both E. coli and coliforms in just 24 hrs.
