Yeast and Mold

Molds is a type of fungus that is micellular and grow filamentous branches called hyphae. Each cell has identical nuclei and they grow in colonies as a single organism. Yeast are single celled microbes are white and thread like whilst molds can be green, orange, black, brown, pink or purple.

Occurrence of Yeast and Molds

Molds tend to grow in damp dark areas whilst yeast can be found everywhere on skin, in stomachs of animals, on berries and plants. Yeast and molds are highly adaptive of their environments and can grow over a wide range of temperatures and pH values. There are about 1000 species of molds and about 1500 species of yeast. Molds reproduce asexually or sexually through spores whilst yeast grows by mitosis or budding. Yeast and molds are responsible for the spoilage of food products. Some molds develop mycotoxins which can cause acute or chronic poisoning. Due to their widespread prevalence the enumeration of yeast and molds is an important parameter for HACCP.

Arasains Indonesia supplies potato dextrose agar from Biolife for the enumeration of yeast and molds. The PDA must be acidified with 2ml of sterile 10% tartaric acid prior to pouring onto petri dishes. Mycological agar and YM 11 Agar with chlorotetracycline are also available.

Film Method - Compact Dry YM

R-Biopharm Indonesia supplies the Compact Dry YM. The ready-to-use plates consist of a special 50 mm diameter petri dish containing a detection specific nutrient pad. During growth on the nutrient medium of Compact Dry YM colonies of yeasts and molds show different color reactions and therefore they are easily to differentiate. The chromogenic substrate X-Phos initiates a blue coloration of nearly all yeast colonies. Molds will form their typical three-dimensional structures in the air space between nutrient pad and plate cover. The coloration of the air structures might be different according to the special type of mold. Growth of bacterial species on Compact Dry YM is inhibited by antibiotics which are added to the medium.

    Biocontrol simplate cannot differentiate between yeast and molds. This is because there is so many species. Differentiation is done by means of morphology. However the enumeration of these is difficult as some grow faster than others.
